Enzymatic Synthesis of Anserine in Skeletal Muscle by N-Methylation of Carnosines

چکیده

Anserine biosynthesis may occur in skeletal muscle by two and possibly three pathways: (a) condensation of l-methylhistidine with p-alanine (1, Z), (6) direct N-methylation of carnosine (3, 4), and, possibly, (c) P-alanyl transfer from carnosine to 1-methylhistidine (5). The relative importance of these various routes is unevaluated, although some studies in viva suggest that species differences are significant, e.g. methionine but not l-methylhistidine is utilized for the synthesis in viva of anserine in the rat, in contrast to the chick which can utilize either compound (7, 8). Winnick and Winnick (3) have shown that S-adenosyl-nmethionine may function as a methyl donor for the synthesis in vitro of anserine and to a lesser degree for synthesis of lmethylhistidine. The present paper describes a further investigation of the properties and specificity of the enzyme obtained from chick muscle, carnosine-N-methyl transferase, which catalyses the synthesis of ‘anserine by transfer of the methyl group of S-adenosylmethionine to carnosine. A preliminary report of this work has appeared (4).

The effect of 8 weeks endurance exercise on the content of total and phosphorylated AKT1, mTOR, P70S6K1 and 4E-BP1 in skeletal muscle FHL of rats with type 2 diabetes

Introduction: The mTOR pathway in skeletal muscle plays a very important role in the protein synthesis process, which plays a very important role in proteins. The role of endurance exercise has not yet been studied in this important pathway in protein synthesis in people with type 2 diabetes. The purpose of the present study was to investigate the effect of 8 weeks endurance training on the con...

Comparison of the Alterations of Gene Expression Related to Signaling Pathways of Synthesis and Degradation of Skeletal Muscle Protein Induced by Two Exercise Training Protocols

Background and Objectives: Skeletal muscle mass depends on the balance between synthesis and degradation of muscle protein, which changes with aging and disease. The aim of the present reserch was to examine the effects of two exercise training protocols on alterations of some genes involved in pathways of protein synthesis and degradation in order to achieve a more effective training program i...

Carnosine and Anserine in Mammalian Skeletal Muscle *

Two imidazole derivatives, carnosine and anserine, have been isolated from vertebrate skeletal muscle. Carnosine may be readily obtained from the ox (l), horse (2), pig (3), etc. Anserine, a methyl carnosine, has been isolated from birds, a reptile (4), fish (5), and certain mammals (6). The presence of the two compounds in muscles from the same animal has been adequately demonstrated only once...

Intracellular Distribution of Carnosine and Anserine in Skeletal Muscle.

The experiments of Bach and Langley (1) with frog muscle suspensions indicated that the carnosine of this tissue was freely diffusible upon dialysis of homogenates. On the other hand, Reddy and Hegsted (2) observed that the “debris” of rat skeletal muscle homogenates contained 43% of the total carnosine. This debris, representing material sedimented at 650 x g, still retained half of its initia...
